We describe the development of a novel technique for single-molecule fluorescence tracking of biomolecules in three-dimensions inside living cells. This novel technique was applied to the study of molecular motors belonging to the Class V of the Myosin Family. We studied Myosin Vb from a single-molecule point of view for the first time and we will investigated its role in long term potentiation in neurons through imaging inside living neurons thanks to he novel technique that we developed during this thesis.
