Correlation between production parameters and biochar quality. Evaluation of mutagenic potential of biochar organic extracts by bacterial mutagenicity test (Ames test). Biochar engineering with selected PGPM microorganisms and evaluation of the fertilizer potential of this new association. Study of the effects deriving from the use of biochar, biochar associated with the microbial consortium created with the selected strains and from the use of only microbial consortium on the growth in pots and on the quality of two tomato cultivars: Heinz 3402 and Ailsa Craig. The parameters observed were physiological (fresh and dry biomass, shoot height and root length, number of leaflets, leaf area, water content) and phytochemical (chlorophyll, carotenoids, pheophytins, antioxidants by anthocyanins and enzymatic catalase activity, hydrogen peroxide content). In addition, metabolomics has been evaluated in three different organs for each plant (leaves, stems, roots) considering 6 main classes of metabolites: antioxidants, organic acids, amino acids, fatty acids, sugars and nucleotides/sides/bases. Finally, the effects on the soil microbial community were studied using the Community-Level Physiological Profiling (CLPP) technique by analyzing the soil of plants exposed to experimental treatments.
Caratterizzazione delle proprietà chimiche, fisiche e biologiche di tre campioni di biochar di origine vegetale e prodotti mediante gassificazione a diverse temperature. Correlazione tra parametri di produzione e qualità del biochar. Valutazione del potenziale mutageno di estratti organici di biochar tramite test batterico di genotossicità (test di Ames). Ingegnerizzazione del biochar con microorganismi PGPM selezionati e valutazione del potenziale fertilizzante di tale associazione. Studio degli effetti derivanti dall'utilizzo di biochar, di biochar associato al consorzio microbico creato con i ceppi selezionati e del solo consorzio microbico sulla crescita in vaso e sulla qualità di due cultivar di pomodoro: Heinz 3402 e Ailsa Craig. I parametri osservati sono stati quelli fisiologici (biomassa fresca e secca, altezza del fusto e lunghezza delle radici, numero di foglie, area fogliare, contenuto idrico) e fitochimici (contenuto di clorofilla, carotenoidi, feofitine, antiossidanti mediante antocianine e attività enzimatica della catalasi, contenuto di perossido di idrogeno). Inoltre, la metabolomica è stata valutata in tre organi diversi per ciascuna pianta (foglie, fusti, radici) considerando 6 classi principali di metaboliti: antiossidanti, acidi organici, amino acidi, acidi grassi, zuccheri e nucleotidi/sidi/basi. Infine, gli effetti sulla comunità microbica del suolo sono stati studiati mediante la tecnica del Community-Level Physiological Profiling (CLPP) analizzando i suoli delle piante esposte ai trattamenti sperimentali.
