The present study evaluated the percentage changes of lymphocyte subpopulations in the peripheral blood of bitches undergoing elective or during pthology ovariohysterectomy surgery (pyometra in these cases) and the presence and localization of the same cellular subpopulations in the uterine environment by immunohistochemical examination. Initially, the general framework of development and maturation of the lymphocyte subpopulations considered is described, followed by a review of the literature regarding the percentage changes of the same subpopulations in both veterinary and human medicine. Next, the anatomy of the reproductive system of the bitch and the estrous cycle are briefly described. In addition, the principles of flow cytometry are also exposed. The blood profile of 25 bitches of different breeds, sizes and ages was investigated. The lymphocyte subpopulations examined were B lymphocytes with marker CD20 and CD21, T lymphocytes with markers CD3, CD4, CD8, TCRγδ, and regulatory T lymphocytes (Tregs). The same cell populations were considered on the excised uteri by immunohistochemistry. Cytofluorometric analyses were performed in three stages: in the first stage, all subjects were considered without distinction; in the second stage, subjects were divided into healthy and with pathology (19 and 6 subjects, respectively); and in the third stage, subjects were divided according to age (5 young, 15 adult, and 5 elderly subjects). In the first analysis, statistically significant values were obtained at %lymphocyte/PBMC, CD3+, CD8+, and CD21+; in the second stage of analysis, statistically significant values were obtained at %lymphocyte/PBMC and CD4+; in the third stage, statistically significant values were at %lymphocyte/PBMC, CD8+, and CD21+. For what concerns immunohistochemical examination, healthy uteri were compared with pathological uteri (undergoing pyometra in these cases), obtaining statistically significant values at the CD3+, CD8+ and CD20+ populations. Data obtained were subjected to statistical analysis and then compared with the results obtained by other authors.
Il presente studio ha valutato le modificazioni percentuali delle sottopopolazioni linfocitarie nel sangue periferico di cagne sottoposte a intervento di ovarioisterectomia elettivo o in corso di patologia (in questi casi piometra) e la presenza e localizzazione delle stesse sottopopolazioni cellulari in ambito uterino attraverso l’esame immunoistochimico. Inizialmente viene descritto il quadro generale di sviluppo e maturazione delle sottopopolazioni linfocitarie considerate, seguito da una analisi della letteratura riguardante le modificazioni percentuali delle stesse sottopopolazioni sia in medicina veterinaria che in medicina umana. Successivamente, viene descritta in breve l’anatomia dell’apparato riproduttore della cagna e il ciclo estrale. Inoltre, vengono anche esposti i principi di citometria a flusso. È stato indagato il profilo ematico di 25 cagne di diverse razze, taglia ed età. Le sottopopolazioni linfocitarie prese in esame sono i linfociti B con il marcatore CD20 e il CD21, i linfociti T con i marcatori CD3, CD4, CD8, TCRγδ e linfociti T regolatori (Tregs). Le stesse popolazioni cellulari sono state prese in considerazione sugli uteri asportati tramite immunoistochimica. Le analisi citofluorimetiche sono state eseguite in tre fasi: nella prima fase sono stati considerati tutti i soggetti senza distinzione, nella seconda i soggetti sono stati divisi in sani e con patologia (rispettivamente 19 e 6 soggetti), mentre in terza battuta sono stati suddivisi in base all’età (5 soggetti giovani, 15 adulti e 5 anziani). Nella prima analisi sono stati ottenuti valori statisticamente significativi a carico della %linfociti/PBMC, CD3+, CD8+ e CD21+; nella seconda fase di analisi sono stati ottenuti valori statisticamente significativi a carico di %linfociti/PBMC e CD4+; nella terza fase invece i valori statisticamente significativi riguardavano la % linfociti/PBMC, CD8+ e CD21+. Per quanto riguarda l’esame immunoistochimico sono stati messi a confronto uteri sani con uteri patologici (in corso di piometra in questi casi), ottenendo valori statisticamente significativi a carico delle popolazioni CD3+, CD8+ e CD20+. I dati ottenuti sono stati sottoposti ad analisi statistica e successivamente confrontati con i risultati ottenuti da altri autori.
