Abstract Proof of concept: As well as reporter above, we thought to coin the acronym as “Prometheus” as representative words of “PRotein Molecular Expression for THErapeutic USefulness”. In fact, looking in the proposed sentence and the significant concept of Prometheus history, this acronym seems to be considered a “nomen omen” proposition. Indeed, looking for the protein expression, we can assess that might be represent the “final cut” of nucleic acid featuring protein expression inside the triage of biomolecules. Nevertheless, the correct both validation and quantification of protein expression, might represent the final indication from the bench to bed side connection. Furthermore, according the value and the role of quantified marker, it’s expression can be associated with different field of medicine, including: clinical treatments, diagnosis and validation systems. Biomarker definition: It is usually possible to divide the “molecular markers” into three different groups, according to their function in relation to their application in clinical practice. In fact, we can define the marker as follows: Diagnostic, Predictive and Prognostic which seek their importance in the diagnosis, clinical treatment and outcome of patients, respectively. In particular, we would like to focus our attention on the protein expression of molecular markers. Aim: The aim of this thesis is to defend the usefulness of protein quantification in different systems by pursuing different goals. So far, included in this work, we would like to present 4 publications which we believe can represent and defend the following list of arguments: 1)The concept of the project;2) The technical approaches used for protein quantification; 3)New attempts at protein quantification; 4) The value of protein expression both within the clinic and in medical research. The following four articles are reported in their respective chapters.
