It has been demonstrated that adhesion can control the balance between cell survival and apoptosis (Gilmore, Owens et al. 2009), or that there is a strong association between cell shape and its commitment to a differentiation lineage . The goal of the present report was therefore to study the behavior of murine cell lines and primary cells from bone marrow, when cultured on commercially pure titanium surfaces in the absence or in the presence of a novel phospho-serine/poly-lysine biocoating, and investigate whether it could positively affect cell behavior. The murine osteoblastic cell line MC3T3 was plated on smooth (polished) titanium discs, sand-blasted, acid etched (SAE) titanium discs and SAE discs with biocoating (SAE+Co) and cultured in complete medium enriched with ascorbic acid for up to 6 days. Cell adhesion, as measured by MTT assay, was improved on SAE+Co when compared to the other groups at 48 hours. When we measured the expression of differentiation genes, we observed, from day 3 to day 6 of culture, an increase in the levels of transcripts of Alkaline Phosphatase and Osteocalcin, two markers of osteoblastic phenotype. This increase was higher in cells growing on SAE+Co surfaces. The expression levels of Osterix, an early osteoblastic marker decreased from day 3 to day 6 in all groups, but at day 6 they were higher on SAE+Co.
