Apaf1 (Apoptosis protease activating factor) has a prominent function in the mitochondrial pathway of apoptosis, since it forms the apoptosome, a multimeric complex that activates the initiator caspase-9. It is involved in developmental apoptosis, in cancer and also in other cell functions independently of apoptosis. Although Apaf1 plays these important roles, its regulation is still far from clear. We analysed its promoter with the aim of identifying transcriptional factors able to regulate its activity. We analysed the sequence in silico, looking for putative binding sites recognized by transcriptional factors. In order to verify the role of these factors in Apaf1 gene regulation, we experimentally tested some of them for their ability in modulating the murine Apaf1 promoter activity. We also generated several deletion constructs with the aim of identifying important regions for the transcriptional regulation. In particular we analysed the effect of the transcriptional factor Pax4 on Apaf1 gene, observing that it causes an increase in Apaf1 promoter activity. We were also able to identify a cisacting region of 50bp, which negatively regulates Apaf1 promoter activity.
Apaf1 svolge una funzione rilevante nella via apoptotica mitocondriale poiché forma l’apoptosoma, un complesso multimerico che attiva la caspasi iniziatrice 9. La proteina Apaf1 è coinvolta nell’apoptosi durante lo sviluppo, nel cancro e anche in altre funzioni indipendenti dall’apoptosi. Sebbene Apaf1 svolga questi ruoli importanti, la sua regolazione è ben lontana dall’essere chiarita. Abbiamo analizzato il promotore con lo scopo di identificare dei fattori trascrizionali capaci di regolarne l’attività. Abbiamo analizzato la sequenza in silico, cercando dei possibili siti di legame riconosciuti da fattori trascrizionali. Per verificare il ruolo di questi fattori nella regolazione del gene Apaf1, ne abbiamo testati alcuni sperimentalmente per la capacità di modulare l’attività del promotore murino di Apaf1. Inoltre abbiamo prodotto per delezione diverse costruzioni geniche con lo scopo di identificare delle regioni importanti per la regolazione trascrizionale. In particolare abbiamo analizzato l’effetto del fattore trascrizionale Pax4 sul gene Apaf1, osservando che questo causa un aumento dell’attività del promotore. Inoltre siamo stati in grado di identificare una regione di 50 bp, agente in cis, che regola negativamente l’attività del promotore Apaf1.
